The Blood Flower quest is the gate between you and Isolation Island, the area added in Sailor Piece's Ghoul update. Six flowers are scattered across Sea 2, most of them tucked behind cliffs, inside crevices, or hidden near boss arenas. The quest itself isn't hard once you know where to look, but without a map it turns into a frustrating 30-minute wander. Here's every location, in the order that makes the most sense to run them.
How do you start the Blood Flower quest?
Travel to the Sea 2 Starter Island and find the mountain range sitting just behind the pack of Delinquent enemies near the Spawn Crystal. Climb to the top of these mountains and you'll find the Blood Flower NPC waiting up there. Speak to him and he'll task you with collecting six Blood Flowers spread across the world. Once all six are in hand, the quest completes instantly and Isolation Island unlocks.

Below is every flower location at a glance, followed by detailed directions for each one.
Blood Flower 1: Sea 2 Starter Island
After stepping through the teleport portal on Sea 2 Starter Island, turn left and look at the cliffs running along that side of the island. Climb them and you'll find the first Blood Moon Flower sitting between some rocks, facing out toward the ocean. Hard to miss once you're at the right elevation.

Blood Flower 2: Bizarre Island
Head to Bizarre Island and make your way to the coliseum. Go behind it and hop over the cliffs at the rear. Drop down to the bottom of the cliff face and look into the crevice there. The second Blood Moon Flower is wedged inside it. This one sits right at the base of the cliff, so don't stop at the top.

Blood Flower 3: Punch Island
This one is in the southwest corner of Punch Island. Find the NPC who hands out elimination quests for the nearby Fast Ninja mob pack, then look at the skyscrapers directly opposite him. Climb those buildings and find the one with an Angel Halo on top. Drop down the far side of that building and the third Blood Moon Flower is on the ground waiting for you. This is also the same spot where you find the halo for the Archangel Race, so if you've done that before, the building will look familiar.
The Angel Halo on top of the correct skyscraper is a reliable visual marker. Scan the rooftops as you climb and you'll spot it quickly.
Blood Flower 4: World Island Sea 2
Spawn into World Island Sea 2 and drop down from the sky island to the lower island beneath it, where the traders are set up. Head to the northeast corner and find the Spec Passive and Power traders. The fourth Blood Moon Flower is hiding behind the trees just to their left, close to the water's edge.
Blood Flower 5: Slayer Island
This is the trickiest one. On Slayer Island, navigate to the Sun God boss arena and walk toward the house at the back of it. From there, face right and move forward in a straight line, climbing up the rocks and cliffs ahead. Keep going until you reach a point where there's a drop to your right. Drop into it and check the bushes near the tree at the bottom. The fifth Blood Moon Flower is tucked in there.
Blood Flower 6: Blue Planet
The final flower is at Blue Planet, near the spaceship. Find the Goku NPC (listed in-game as the Empyrean Trainer) just west of the spaceship. Climb over the rocks he's standing in front of and look in the crevice directly behind them. The sixth and final Blood Moon Flower is there.
Don't confuse the Empyrean Trainer's rock formation with other nearby terrain. The flower sits specifically in the crevice behind his rocks, not the wider cliff area around the spaceship.
What do you get for completing the Blood Flower quest?
The moment you collect the sixth flower, the quest finishes automatically. Isolation Island unlocks immediately, no extra steps required. Head to the teleporter and you can travel there straight away.
On Isolation Island you can acquire a new melee style and summon the Black Reaper boss. Grinding the Black Reaper is what most players are actually here for, since it drops the gear tied to the Ghoul update content.
